Pectinase production by fungal-based solid-state fermentation of selected agricultural wastes

Agricultural waste is the residues from growing and processing raw agricultural products. They can be properly managed using fungi to produce economical materials such as pectinase enzymes that act biological agents which are applicable in many areas. This research was aimed at producing by fungal-based solid-state fermentation of wastes. Fungi were isolated dump sites soil pour plate method. These fungal isolates inoculated into pretreated waste, including watermelon, mango, pineapple peels. The degradation period 15 days on mineral salt solid media separate flasks for each fungus. incubated room temperature (28°C). assayed pectin DNSA (dinitrosalicyclic acid). Fungal identified screened production standard methods. pH optimized study effect activities procedures purified column sephadex G-100. Molecular identifications carried out 18srRNA gene primers. Aspergillus niger strain AGF3 (OM273994), Penicillium Chrysogenum AGF2 (OM273982) Cladosporium tenuissimum AGF1 (OM273981). (OM273994) produced highest specific mango (2400 U/mg. protein/ mL it compared what (OM273981) (1590 protein/mL). High pecrtinase activity also obtained peels (2200 protein/mL) tenussimum It concluded this best producers peels, while chrysogenum substantial all three wastes but not higher levels.
